SUMMARY:Milner Therapeutics Institute: Driving innovation and therapeutic discoveries across the ecosystem
DESCRIPTION:🗓️ 31 March ⏰ 3:00-4:00pm 📍Lecture Theatre\, Jeffrey Cheah Biomedical Centre \nThis event is presented as part of the 2026 Cambridge Festival. \nSince its inception in 2016\, the Milner Therapeutics Institute has been a driving force in connecting academia and business in Cambridge and beyond. With a distinctive model that unites therapeutics research\, partnerships\, and enterprise\, the Institute aims to transform pioneering science into life-changing therapies.  \nAt this event\, our Executive Director Professor Cathy Tralau-Stewart will reflect on how Professor Sir Tony Kouzarides turned a bold vision into reality – launching a completely new and unique department that bridged the gap between academia and industry. Cathy will explore how this innovative model compares with other translational research institutes around the world\, highlighting what makes Milner distinct in its approach and impact. She will also chart the Institute’s journey to date\, celebrate key milestones\, and share her perspective on how Milner will continue to drive discovery and innovation in the life sciences. \nRegister at: milner.glueup.com/event/milner-therapeutics-institute-driving-innovation-and-therapeutic-discoveries-across-the-ecosystem-160327/

SUMMARY:FGSL Webinar: Single-cell Intercellular CRISPR Screen Reveals Stromal Regulators of Colorectal Cancer Plasticity
DESCRIPTION:🗓️ 14 April\n⏰ 11:00am-12:00pm\n📍 Online \nJoin the Functional Genomics Screening Laboratory (FGSL) team as we welcome Corinne Molyneux for this special webinar. \nSpeaker: Corinne Molyneux\, University College London\n\nAbstract: Colorectal cancer (CRC) displays high levels of nongenetic phenotypic plasticity\, allowing cancer cells to dynamically shift between different functions\, enabling therapy evasion. This is particularly evident in poor prognosis\, stromal-rich CRC tumours. These tumours also have an elevated presence of revival cancer stem cells (revCSCs). In the microenvironment of these tumours cancer-associated fibroblasts (CAFs) act as dynamic regulators driving the reversible acquisition of YAP-dependant\, slow-cycling\, chemoresistant features that define the revCSC state. The soluble factors that are regulating this transition were however undefined. \nTo resolve this question\, we developed a high-throughput single-cell CRISPR assay to systematically perturb the CAF secretome when in direct co-culture with patient derived organoids (PDO). Individual components of the secretome were targeted by synthetic gRNAs in Cas9 expressing CAFs before co-culture. The perturbed co-cultures were then assessed via multiplexed mass cytometry\, capturing cell fate markers\, post-translational modification (PTM) signalling\, cell-cycle activity\, and apoptosis in both PDOs and CAFs at single-cell resolution. \nUsing this platform we have successfully targeted 202 ligands in our PDO+CAF model and quantified the role of each in the revCSC transition\, unveiling the primary effector of stromal-induced CRC plasticity changes. These findings demonstrate that CRC phenotypic plasticity is regulated by stromal-derived signals\, providing new mechanistic insight into therapy failure and describing a potential therapeutic intervention strategy to overcome plasticity driven chemoresistance. \nRegister at: cam-ac-uk.zoom.us/meeting/register/I4NjVGD2T4Oui4FWDTaJrw#/registration
