SUMMARY:Webinar: Harnessing the power of whole genome arrayed screening to discover new mechanisms and targets in rare genetic conditions
DESCRIPTION:🗓️ 9th October\n⏰ 10:00am\n📍 Online (Zoom) \nJoin us for this webinar presented by the MRC-AstraZeneca-University of Cambridge Joint Functional Genomics Screening Laboratory. \nSpeaker: Dr Delphine Larrieu\, Altos Labs \nAbstract: Progeria syndromes are very rare\, incurable premature aging conditions recapitulating most aging features. They are caused by mutations in nuclear envelope proteins\, who play major roles in maintaining the structure of the nucleus\, as well as the organisation of the chromatin. As a result of these mutations\, patient-derived cells display a dramatic loss of nuclear envelope integrity\, characterised by nuclear shape distortion\, nuclear envelope ruptures and abnormal localisation of nuclear envelope proteins. With the goal of identifying new players that could restore nuclear envelope integrity and cell health in Progeria patient cells\, we designed a whole genome screen\, looking for genes that could reverse multiple nuclear envelope associated phenotypes at once. These phenotypes can only be assessed by microscopy on adherent cells\, preventing the use of pooled\, flow cytometry-based screening strategies. We therefore implemented a whole genome\, CRISPR arrayed screen that proved to be a powerful approach for identifying new genes enhancing cellular resilience in premature aging and uncover novel biology behind progeria-associated cellular dysfunction. \nClick here to register >>

SUMMARY:Webinar: Arrayed CRISPR screening for phenotypic discovery at the MRC-AstraZeneca-University of Cambridge Joint Functional Genomics Screening Laboratory (FGSL)
DESCRIPTION:🗓️ 21st November\n⏰ 4:00-5:00pm\n📍Online \nSpeakers:\nUlrike Künzel\, FGSL Lead\, AstraZeneca\nand\nChun Hao Wong\, FGSL Lead\, Milner Therapeutics Institute\nThe MRC-AstraZeneca-University of Cambridge Joint Functional Genomics Screening Laboratory (FGSL) leverages the unique features of arrayed screening\, whereby individual genes are edited via CRISPR in plate-based format\, to uncover the complexity of developmental and disease signatures using human cellular models. The laboratory – hosted at the Milner Therapeutics Institute\, University of Cambridge – is equipped with a high-throughput screening platform to enable functional endpoint acquisition including high content imaging and flow cytometry. \nThis presentation will showcase previous phenotypic screens in complex cell models\, in which we overcame some of the inherent challenges in delivering CRISPR/Cas9 reagents. We will also present routes to form either academic or small/medium-sized enterprise collaborations with the FGSL for arrayed screening. \nRegister at: insidescientific.com/webinar/arrayed-crispr-screening-for-phenotypic-discovery-at-the-mrc-astrazeneca-ucam-functional-genomics-screening-laboratory-fgsl/

SUMMARY:FGSL Webinar: Genetic modifier screen in human neurons reveals interaction between SARM1 and rare neuropathies
DESCRIPTION:🗓️ 11 April\n⏰ 11:00am-12:00pm\n📍 Online \nJoin the Functional Genomics Screening Laboratory (FGSL) team as we welcome Dr Andrea Elser for this special webinar. \nSpeaker:\nDr. Andrea Elser\, Applied Stem Cell Sciences Team\, Centre for Genomics Research\, Discovery Sciences\, BioPharmaceuticals R&D\, AstraZeneca\, Gothenburg\, Sweden. \nAbstract:\nRare neuropathies\, chemotherapy-induced neurodegeneration and SARM1 activity share common characteristics. NAD-consuming enzyme SARM1 plays a central role in programmed axon death and its loss-of-function has been shown to be protective in subtypes of Charcot-Marie-Tooth disease and Hereditary Spastic Paraplegia. These diseases are characterised by neurite degeneration and mitochondrial defects and currently lack effective therapies. SARM1 inhibitors may provide a much-needed disease-modifying opportunity for certain mechanistic subtypes of rare neurological disorders. To evaluate if SARM1 loss-of-function can protect against neurite degeneration linked to rare neuropathies\, we performed arrayed phenotypic CRISPR-Knock-out (KO) screens in wildtype and SARM1-KO human iPSC-derived neurons\, focusing on 60 disease-associated genes. In this webinar\, we will present in vitro model validation data and how the genetic screen was used to identify rare neuropathies where SARM1 inhibition can be neuroprotective. \nRegister at: cam-ac-uk.zoom.us/meeting/register/1pZA1ZYPQmGL7kA-TP2UZQ#/registration
